If you set this option to mark e-mail as read, this is no longer a preview pane but a viewing pane. You are then left with the option to 'view' mail horzontally, vertically or in a new window.

 
My preference is to mark the e-mail as read by default but make sure the UI uses consistent presentation and concepts.

Off by default, so that clicking on the wrong msg doesn't mark it read; so that using arrow keys to traverse a view doesn't mark bunches of docs read; and because a quick scan doesn't mean you read something necessarily. Most folks I know don't really read their e-mail in the preview pane--they're just previewing it (sorry to be redundant, but that's is what it is), more like an extended subject. ;-)

 
I for one use unread marks for e-mail management; skimming a preview doesn't mean I'm done with that e-mail. (Yes, I use the Insert key toggle plenty. ;-)
 
I hate Apple Mail because preview pane = anything I click on is read. Very annoying, constantly having to mark things as unread.
 
All that said, I stopped using preview pane in mail when it started messing up the frames and my mail would open in two windows, one frames-without-content and the other content-without-the-frames. (That bug might've been fixed already, but I got out of the habit of using preview.)

for myself I like to turn it on but I think that for the majority of my users it needs to be off because it is a common habit to use unread marks to indicate messages that need to be dealt with later.

The time delay in Outlook before it marks messages as read is the insurance against Kendall's concern about accidentally marking messages as read

 
The 5 second time delay that Brian noted in Outlook before it marks messages as read is actually adjustable. I found the refresh of the screen while I was reading the message to be so distracting that I set the delay to the minimum, which is zero (correcting my statement above that it is 1 second).
 
I used to use unread marks to track whether I was "done" with an email, but now I use follow-up flags in both Notes and Outlook. So now I have more information at a glance. I know whether I've looked at it at all, and I know whether I'm done.

I agree with Kendall that the default should be OFF. I think most users don't want return receipts or unread marks to be triggered when they preview an email. If you're a keyboard user, you may be scrolling down with the arrow key and stop at an email. That's doesn't mean you read it. It could just mean that's where you were last when you stopped to answer the phone.

 
I also found it annoying how some email programs mark emails read after a time delay. You notice it while you're previewing the email and it's distracting

I often scan through unreads using preview (note the word 'preview' i.e. not actioning) to see if there is anything important, before deleting. I then want the option to go back on some when I have chance to deal with them in detail. I don't want to then have to start adding flags, or marking back as unread. That said, as long as it is switchable I suppose it doesn't matter.
 
And please don't base on what Outlook does, do what is best. There is already too much being sacrificed to emulate Outlook.

in general, my users do not expect a document to turn "read" when it is previewed, as well as they wish a return receipt shoud not be sent ...

I use to scan unread with tabs, leaving unread if I still need time to manage, and use the 'ins' key to toggle to read status.
 
On the other side, it looks that the status of preview panel is not retained any more (when restarting notes).Bug or feature?

ONI can't remember the last time I opened a separate window to view an email.

Needs a timed setting like Outlook. If you happen to use the arrow keys in your Inbox, you inadvertantly mark messages read that aren't. I vote off by default.

I use mail preview. Most of the mails I read are never properly opened. I think the "Mark Documents 'Read' when Opened in Preview" option should be On by default.
